Quantitative Model for Reversibly Photoswitchable Sensors

Composed of a reversibly photoswitchable unit allosterically linked to sensing module, sensors (rs-sensors) represent new and attractive strategy quantitatively read-out analyte concentrations. However, their kinetic response illumination is complex, much attention required from the design application steps. Here, we exploit generic model rs-sensors which enables us point key thermokinetic parameters, such as dissociation constants rates for exchange toward analyte, cross-sections photoswitching. The allows evaluate robustness analyzed parameters introduce methodology reliable use. Model have been experimentally tested on newly reported calcium sensor based green fluorescent protein calcium-sensing module integrating calmodulin an RS20 peptide.
